The largest site I know of is http://www.boston.com, the website of the
Boston Globe (owned by the New York Times).
It is one of the largest websites in New England, and they converted
recently from a commercial/proprietary CMS to Zope.
Marc Lavallee of Boston.com gave a presentation on their experience at
the third international OSCOM meeting at Harvard Law School Berkman
Center for Internet and Society.
